چکیده

Mucin 1 (MUC1) is a type I transmembrance protein that consists of an extracellular domain containing a variable number of tandem repeats (VNTRs), a transmembrance domain and a cytoplasmic domain (Gendler., 2001; Schuman et al., 2005). The 20-amino acid sequence (GVTSAPDTRPAPGSTAPPAH) repeats of the VNTRs of MUC1 are heavily glycosylated (Gendler et al., 1995). While MUC1 is expressed at low levels on the apical surface of ductal epithelia cells in many organs, on tumor cells it is overexpressed, under-glycosylated and associated with loss of polarity (Croce et al., 2007; Seregni et al., 1997; Brockhausen et al., 1995). These characteristic changes of MUC1 exposes its protein backbone, thereby making the tumor vulnerable to the immune system. Therefore, MUC1 epitopes are considered potential antitumor targets. Antitumor activity of lentivirus-mediated interleukin -12 gene modified dendritic cells in human lung cancer in vitro.

OBJECTIVES Dendritic cell (DC)-based tumor immunotherapy needs an immunogenic tumor associated antigen (TAA) and an effective approach for its presentation to lymphocytes. In this study we explored whether transduction of DCs with lentiviruses (LVs) expressing the human interleukin-12 gene could stimulate antigen- specific cytotoxic T cells (CTLs) against human lung cancer cells in vitro. MET...

Dendritic cell-glioma fusion activates T lymphocytes by elevating cytotoxic efficiency as an antitumor vaccine

BACKGROUND Hybrid cells produced by fusions of tumor and dendritic cells (DC) have demonstrated remarkable efficacy for priming the anti-tumor immune response. In the current study, we examined the antitumor activity of cytotoxic T lymphocytes (CTLs) primed in response to a tumor vaccine comprising a glioma-DC fusion as part of a therapeutic against glioma. MATERIAL AND METHODS Primary cultur...

Dendritic cells transfected with lentiviral vector-encoding hTERT peptide augment antitumor T cell response in vitro.

HIV-1 derived lentiviral vectors (LVs) have emerged as a powerful tool for gene delivery. hTERT is an ideal tumor-associated antigen with which to develop a potential dendritic cell (DC) vaccine. The purpose of this study was to construct a recombinant lentivirus vector of the hTERT peptide and to determine the hTERT-specific cytotoxic T lymphocyte response elicited by DCs transfected with hTER...

Novel approach to the characterization of melanoma associated-peptide-specific CTL lines from Japanese metastatic melanoma patients.

Melanoma-associated antigens, MART-1, tyrosinase, gp100 and MAGEs, are typical melanoma-specific tumor antigens which can potently induce immune responses in metastatic melanoma patients treated with peptide vaccines. In the present study, we established a dendritic cell (DC)-based HLA-A2 melanoma-associated peptide (MART-1 or gp100)-specific CTL induction method and characterized the CTLs usin...

Generation of kidney cancer-specific antitumor immune responses using peripheral blood monocytes transduced with a recombinant adenovirus encoding carbonic anhydrase 9.

PURPOSE Carbonic anhydrase 9 (CA9) is the most promising molecular marker described for renal cell carcinoma (RCC) to date. We investigated whether transduction of monocytes from peripheral blood with adenovirus encoding the CA9 gene (AdV-CA9) could stimulate a T-cell mediated immune response against cancer cells expressing CA9. The ability to consistently generate a T-cell response is an impor...
